Effect

• Prokinetic agents (D2i, 5HT4A) → increase GI mobility, limited to upper GI (esophagus to proximal jejunum)

  

• Anti-emetic

  

Conditions:

• Gastroparesis → hospitalized patient with acute flare and unable to tolerate oral intake (Domperidone is only PO) → Constipation
• N/V - Nausea/vomitting

CI:

Side effects:

• Neurological symptoms (crosses BBB)
  • Drug-Induced Parkinsonism

Admin:

• IV
• Avoid chronic use

MOA

• increase GI mobility by
  • D2 antagonist: antagonizing D2 receptor → Ach release
  • agonizing 5HT4 receptor → Ach release
• Reduce nausea by
  • agonizing 5HT3 receptor in brain (chemoreceptor trigger zone, medulla)
